چکیده

Recently, therapy with bone marrow mesenchymal stem cells (BMMSCs) has been attempted to relieve rheumatoid arthritis (RA) and reconstruct cartilage injury. However, treatment has been unsuccessful in complete prevention of persistent cartilage destruction and resulted in inferior outcomes of cartilage regeneration. Scaffolds are an important construct in the field of cartilage tissue engineering, but their role in arthritis treatment has not yet been fully examined. Here, we transplanted two types of scaffold-assisted BMMSCs: fibrin geland poly(L-lactide-co-glycolide) ́poly(ethylene glycol) ́poly(L-lactide-co-glycolide) (PLGA ́PEG ́PLGA) hydrogel-assisted BMMSCs referred as FGB and HGB groups, respectively, into subchondral defects for the treatment of antigen-induced arthritis. The administration of exogenous BMMSCs ameliorated joint swelling and decreased both joint surface temperature and inflammatory cytokine levels in both groups. Immune cell composition of the inflammation of surrounding synovium, protection of adjacent cartilage, and improved cartilage repair were also observed. Overall, the HGB group had a better therapeutic efficacy than the FGB group. In conclusion, local transplantation of BMMSCs in subchondral defects presents a novel approach in inducing RA remission and recovery of RA-induced cartilage injury. To induce these changes, the selection of scaffold for cell support is exceedingly important. Further studies are needed regarding the treatment options of subchondral defects in arthritis based on modified scaffold development, application of defined MSCs sources, combination of pharmacotherapeutics, and the addition of factors that inhibit the processes of RA remission, promote the recovery of RA-induced cartilage injury and the relationship between these factors.

P-131: Autologous Transplantation Encapsulated Preantral Follicles into Fibrin Hydrogel Supplemented with Platelet Lysate

Background: Recent improvement in treatments of cancer has significantly increased survival rates of cancer patients. Unfortunately these treatments such as chemotherapy and radiotherapy can sever damage to the ovaries. Ovarian cryopreservation prior to chemo/ radiotherapy and auto-transplantation post-treatment can restore fertility to cancer patients. Hypoxia in the transplant from delayed re...

Use of Mesenchymal Adult Stem Cell for Cartilage Regeneration by Hydrogel

Background and Aims: Cartilage is a very specific tissue, which does not have the capacity to heal and renew itself. Although the invention of the method of surgery with autologous chondrocyte transplantation, developed tools to treat the cartilage lesions, it couldn’t gain a great success due to problems such as damage to the area of donation. Using the mesenchymal stem cells derived from adip...

Co-Transplantation of VEGF-Expressing Human Embryonic Stem Cell Derived Mesenchymal Stem Cells to Enhance Islet Revascularization in Diabetic Nude Mice

Background: Pancreatic islet transplantation has emerged as a promising treatment for type I diabetes. However, its efficacy is severely hampered due to poor islet engraftment and revascularization, which have been resulted to partially loss of transplanted islets. It has been shown that local delivery of vascular endothelial growth factor (VEGF) could accelerate transplanted islet revasculari...

From a Chemical Matrix to Biologically/Biomechanically-Defined Matrices-Optimizing/Correlating Growth Rate and Differentiation Potential of Human Adipose-Derived Mesenchymal Stem Cells

Use of Adipose Stem Cells (ADSCs), obtained easily in a relatively less invasive manner (abdominoplasty) and characterized by flow cytometry, is a classical approach in stem cell research and clinical aspects. Other techniques such as isolation of the cells from bone marrow aspirates  (1) are rather more invasive. Further, it is pertinent to point out that growth rate, differentiatio...

Experimental study on healing of long bone defects treated with fibrin membrane enriched with platelet growth factors and periosteal mesenchymal stem cells in rabbit: radiographical and histopathological evaluations

The present study was designed to evaluate the effects of platelet growth factors and periosteal mesenchymal stem cells on bone healing process, radiographically. Forty male White New Zealand rabbits in five equal groups were used in this study. A 2 mm full thickness bone defect was made in left radial bone of each animal. In group A (control) the defect was left with no medical intervention. I...
